HPLC separation and indirect ultraviolet detection of phosphorylated sugars.

An HPLC method for the separation and analysis of phosphorylated sugars is presented. Ion-exchange chromatography coupled to indirect ultraviolet detection has produced good resolution and sensitivity. Fructose 6-P, glucose 6-P, ribose 5-P, 3-phosphoglyceric acid, ribulose 1,5-P(2), fructose 1,6-P(2), and sedoheptulose 1,7-P(2) can be separated at a sensitivity down to 10 nanomoles. The system ...

Hplc Separation of Phospholipids

The principles involved in the selection of solvents and the use of counter ions, together with the choice of column, for the separation of phospholipids by highperformance liquid chromatography (HPLC) are described.

The oxidation of phosphorylated and non-phosphorylated sugars by mammalian liver.

In 1928 Miiller (1) reported the presence of a new enzyme, a glucose oxidase, in press-juice of Aspergillus niger. He subsequently suggested that it might be a dehydrogenase (2) ; he later showed that it catalyzed the oxidation of glucose to gluconic acid (3). Harrison in 1931 obtained glucose dehydrogenase from acetone-dried mammalian liver by saturating a water extract with ammonium sulfate (...

Comparison of different extraction and detection methods for sugars using amino-bonded phase HPLC.

In this study, different methods are compared in order to quantitate individual sugars extracted from apple samples using methanol and water. Glucose, fructose, and sucrose are separated in 20 min using an amino-bonded carbohydrate column and a 75:25 acetonitrile-water mobile phase, followed by UV (190 nm) and refractive index detection. Variations in the sugar profiles are observed using diffe...

Effect of Capillary Rinsing Protocol on the Reproducibility of Separation in Capillary Electrophoresis with Indirect UV Detection

Capillary electrophoresis (CE) with indirect UV detection is an interesting analytical method for the analysis of drugs and pharmaceuticals. Good and reproducible capillary quality is needed to develop robust methods and to facilitate method transfer in CE. It is widely accepted that preconditioning procedures are indispensable in capillary electrophoresis in order to achieve reproducibility of...
